چکیده
Background: Choriocarcinoma is a malignant trophoblastic tumor that can degrade the uterine basement membrane to facilitate local and distant metastasis. Matrix metalloproteinase-2 (MMP-2) vital proteinase produced by trophoblasts cleave type IV collagen in of epithelium. In addition, E-cadherin controls cell adhesion, which associated with tumorprogression, loss function Vitamin A analogues, including all-trans retinoic acid (ATRA), exhibit anticancer properties, metastasis inhibition. This study aimed investigate ability ATRA prevent trophoblast invasion choriocarcinoma assessing MMP-2 activity. Methods: This administered various doses BeWo (American Type Culture Collection CCL-98) line assessed activity through flow cytometry. The research was performed at Biomedical Laboratory Faculty Medicine Universitas Brawijaya Malang, East Java, Indonesia. Results: cells were exposed 0, 50, 100, 200, 400, or 800 μg/mL ATRA. downregulated after treatment. greatest reduction level observed 200 treatment, but this result not statistically significant (P = 0.550). However, treatment significantly reduced number low levels 0.012). Conclusion: downregulates upregulates E-cadherin. Further vitro animal studies are required evaluate inhibit Bangladesh Journal Medical Science Vol. 22 No. 02 April’23 Page : 336-340

All-trans-retinoic acid: measurement
A quantitative determination of physiological levels of all-trans-retinoic acid (vitamin A acid) in human serum has been developed. A double-phase extraction of 3.5 ml of serum followed by a specific and sensitive high performance liquid chromatography procedure allowed measurement of levels down to 1 ng/ml. Serum concentrations in 37 fasting volunteers ranged from 2.7 to 4.2 ng/ml and fitted a...

Studies on Multifunctional Effect of All-Trans Retinoic Acid (ATRA) on Matrix Metalloproteinase-2 (MMP-2) and Its Regulatory Molecules in Human Breast Cancer Cells (MCF-7)
Background. Vitamin A derivative all-trans retinoic acid (ATRA) is considered as a potent chemotherapeutic drug for its capability of regulating cell growth and differentiation. We studied the effect of ATRA on MMP-2 in MCF-7, human breast cancer cells, and the probable signaling pathways which are affected by ATRA on regulating pro-MMP-2 activity and expression. Methods. Gelatin zymography, RT...
متن کاملAll-Trans Retinoic Acid Increases Aquaporin 3 Expression in Human Vaginal Epithelial Cells
INTRODUCTION Water channel aquaporin 3 (AQP3) is an aquaglyceroporin that transports small neutral solutes and water. All-trans retinoic acid (ATRA), a member of the retinoid drug class, acts as a regulator in several biological processes. AIM To investigate the effect of ATRA on the expression of AQP3 in human vaginal epithelial cells. METHODS Human vaginal mucosal epithelial cells (CRL261...
